dc.description.abstractThe purpose of this study is to determine the extent of viral shedding from the vaccination site following vaccination with a newly Food and Drug Administration (FDA)-licensed smallpox vaccine, ACAM2000. Viral shedding will be determined by collecting lesion, blood, and throat swab samples from volunteers receiving the vaccine and quantifying the total virus present by real-time polymerase chain reaction and plaque assay. In addition. we will assess the ability of povidone iodine ointment (PIO) to reduce viral shedding at the vaccination site, and determine what effect, if any, PIO treatment has on the humoral immune response. Humoral responses of both P10-treated and untreated volunteers will be assessed using a vaccinia virus-specific indirect enzyme-linked immunosobant assay (ELISA) and a plaque-reduction neutralization test (PRNT). The data obtained from this study will be used to determine the effectiveness of PIO treatment for the reduction of viral shedding following vaccination with ACAM2000.
